You shouldn't run it off of a usb but sure. It's no different after compiling it yourself aside from the fact that you have to download a portable version of mysql and all that.

I've used a repack before but I know it's something you shouldn't really do, but it was nice cause it ran off a USB easy. Could take my progress from computer to computer (just playing solo, no networking)

Yes, they add a portable version of mysql (which you can download yourself from mysqls own website) with the compiled binaries (which is what you compile yourself) and the required dlls (which you also have)

Just keep in mind that running it off of a USB drive, not a regular HDD acting as a USB drive but an actual thumbdrive, will deterioate it a lot quicker when you're constantly reading from it and writing to it. You might not be affected by it if the read/write isn't significant enough but they're not built for it.
