It might be set to bind only to 127.0.0.1 and it won't accept connections on another IP-address.

If it's running on the same system, there's no need to change it.

If it was running on a system on the same network, you should use the local IP. There's never any reason to allow connections from the outside, avoid port forwarding 3306 too.
