Source folder - E:\S
CMake Build Folder - E:\B
CMakePrefix folder (copy built files from VS to) - E:\T
Do I need to edit files in the Source directory? Or can I use the VS solution that CMake creates in the Build directory?

You can use vs solution from build directory. Make sure to enable dynamic linking
