If I spawn an ore/herb node, will it respawn fine when gathered or will it mess with any zone pooling for ores/herbs? Can I increase this value (amount of ores spawned per zone) if so?

It'll respawn fine but it won't be in a pool with the other ores. It'll respawn whenever the time is up no matter how many other nodes are nearby.
